Transplantrejection,liketolerance,isaTcell-dependentevent.ThereiscompellingevidencetosuggestthatinductionoftransplanttoleranceisanactivelylearnedprocessinwhichTcellsneedtoengagewiththealloantigensinordertolearntotoleratetheallograft.AfamilyofcytokineswhosereceptorsusethesameIL-2receptorγcchain(alsocalledthecommonγc)playsanimportantroleinregulatingmultipleaspectsoftheallograftresponse(I.e.Rejectionvs.Tolerance).ItisundeniablethatγccytokinescandriveclonalexpansionandeffectormaturationofalloreactiveTcells,andtherefore,targetingsuchcytokinesortheirreceptorcomponentsremainsanattractivewayofblockingtransplantrejection.However,wejuststartedtoappreciatethatγccytokinesalsoregulatetheacquisitionoftransplanttoleranceviaprogrammingactivatedTcellsforapoptoticcelldeathandviaguidingtheevolutionofregulatoryTcells.Thus,understandingpreciselytheroleofγccytokinesinregulatingTcellhomeostasisandTcellregulationiscriticallyimportantintheinductionoftransplanttolerance.
