摘要
Direct-methodphaseextensionhasbeenappliedtotwo-dimensionalelectrondiffractiondataoftheproteinstreptavidin.Structure-factoramplitudesfromelectrondiffractionwerecombinedwithphasesfromthecorrespondingelectronmicrographs.Maximum-entropydiscriminationandclusteranalysiswereusedtoderiveasolutionfromalargenumberofrandomtrials.Thephaseextensionfrom0.3to0.25nmledtosubstantialimprovementofthereconstructedprojectionimagequality.
