ThepurposeofthisstudywastotesttheeffectivenessofsourcevirusstrainforthemanufactureoftheinactivatedSARSvirusvaccine,andestablishanexperimentalmethodandpreliminarystandardforpotencyevaluation.MiceweredividedintogroupsforbeingimmunizedwithcorrespondingseriallydilutedexperimentalSARSvirusinactivatedvaccine.Andtherabbitswereimmunizedwithundilutedvaccine.ChallengeassaywasconductedwithaheterologousSARSvirus.Andtheneutralizationantibodywasdeterminedwithplaquereductionneutralizationtest(PRNT),towhichtheneutralizationantibodyintheconvalescentserumofSARSpatientswascompared.Theexperimentalvaccineviralstrainswereprovedtobesuitableformanufacturingthevaccine.Miceimmunizedbyvaccinesofserialdilutionswereabletoelicitneutralizingantibody.Theantibodytiterfrommiceimmunizedwiththeundilutedvaccinecouldreachupto1:495.2,whilethoseofrabbitsimmunizedwiththeundilutedvaccinecouldreachaGMTof55.0-79.9.ThecapabilityoftheantibodytoneutralizethevirusfromGuangdongismoreefficientthanthatfromBeijing.TheGMTofneutralizingantibodyinSARSconvalescentslivinginsouthandnorthChinarangedfrom50.12to54.95,andthetitersofconvalescentsfromnorthChinawerehigherthanthosefromsouthChina.Miceandrabbitsusedasthemodelforevaluationofpotencyareofsensitivity,andthetestisofreproducibility.Thecandidatechallengeviralstrainsshowedarelativelyconsistenteffectonevaluatingantibodiesproducedbyvariousbatchesanddifferentvaccine-sourcestrains,hencetheycanbeusedtoevaluatepotencyofthevaccine.Themethodfortestingthevaccinepotencyandtheevaluationstandardwasestablishedpreliminarily.
