  • 简介:Itisimportanttounderstandseasonalheavymetalaccumulationindifferentpartsofplantsinordertodevelopthebestphytoremediationpracticesforcontaminatedsoils.Forthispurposeweexposed,1yearoldA.victoriaeseedlingstoZnSO4in4differentconcentrations:0,50,250and500mgZnL-1for45daysovertwogrowingseasons.Subsequently,bioaccumulationofZnindifferentplanttissues(roots,shootsandleafs)wasassessedbyAtomicAbsorptionSpectroscopy(AAS)fortwoperiods.Inaddition,variousgrowthattributes(drybiomass,shootandrootlengths,plantappearance)andfunctionaltraits(leafarea,chlorophylla,bandtotal)weremeasured.TheaccumulationofZnwasinfluencedbytheZnconcentrationinthegrowthmediumandthenumberofgrowingseasons.TheamountsofZnconcentratedintheroottissuesmightindicateA.victoriaeasagoodoptionforphytostabilizationofsoilscontaminatedbyZn.WerecommendthatifA.victoriaeisusedforphytoextractionpurposes,thenitshouldbeharvestedattheendofthefirstgrowingseason(fall)becauseatthistimetheconcentrationsofZnintheabove-groundpartswillbemaximal.

  • 简介:Exogenousapplicationofspermidine(Spd)hasbeenreportedtomodulatephysiologicalprocessesandalleviatesalt-induceddamagetogrowthandproductivityofseveralplantsincludingrice.Employingaproteomicapproach,weaimedatidentifyingriceleafandgrainproteinsdifferentiallyexpressingundersaltstress,andinresponsetoSpdpriortoNaCltreatment.Atotalof9and20differentiallyexpressedproteinspotswereidentifiedintheleavesofsalt-tolerant(Pokkali)andsaltsensitive(KDML105)ricecultivars,respectively.Differentialproteinscommontobothcultivarsincludedaphotosyntheticlightreactionprotein(oxygen-evolvingcomplexprotein1),enzymesofCalvincycleandglycolysis(fructose-bisphosphatealdolaseandtriose-phosphateisomerase),malatedehydrogenase,superoxidedismutaseandahypotheticalprotein(OsI18213).MostproteinswerepresentathigherintensitiesinPokkalileaves.Thephotosyntheticoxygen-evolvingenhancerprotein2wasdetectedonlyinPokkaliandwasup-regulatedbysalt-stressandfurtherenhancedbySpdtreatment.AllthreespotsidentifiedassuperoxidedismutaseinKDML105wereup-regulatedbyNaClbutdown-regulatedwhentreatedwithSpdpriortoNaCl,indicatingthatSpdacteddirectlyasantioxidants.Importantdifferentialstressproteinsdetectedinmaturegrainsofbothricecultivarswerelateembryogenesisabundantproteinswithprotectiverolesandanantioxidantprotein,1-Cys-peroxiredoxin.HighersalttoleranceofPokkalipartlyresultedfromhigherintensitiesandmoreresponsivenessoftheproteinsrelatingtophotosynthesislightreactions,energymetabolism,antioxidantenzymesintheleaves,andstressproteinswithprotectiverolesinthegrains.

  • 简介:Whitebackedplanthopper(WBPH)-resistanceinajaponica/indicadoubledhaploid(DH)ricepopulationestablishedfromacrossbetweenWBPH-resistantjaponicaChunjiang06andsusceptibleindicaTN1,wascomparativelyevaluatedthroughafieldexperimentbasedontheWBPHimmigrantdensityandstandardizedseedboxscreeningtest(SSST).AllthesusceptibleDHlinesinthefieldexperimentbehavedaccordinglyinSSST.However,35ofresistant66lines(53%)inthefield,werecategorizedtosusceptiblegroupsinSSST.Likewise,therewerenosignificantdifferencesinWBPHimmigrantdensitiesamong70DHlinesthatwerehighlyresistanttosusceptibleinSSST.TheresultsrevealedthatSSSTcouldnotevaluateproperlyWBPHresistanceintheDHlines.FourQTLsforWBPH-resistancephenotypedbytheimmigrantdensityweredetectedonchromosomes2,3,4,and11.Ofthem,theQTLonchromosome4wasthemosteffective(LOD21.8,variance78%).FiveQTLsassociatedwithseedlingmortalityweremappedonchromosomes2,3,4,5and6.InadditiontotheQTL(LOD10.5,variance68%)onchromosome4,therewasanothermajorQTL(LOD12.7,variance71%)locatedonchromosome5,whichwasSSST-specificbutmightbeirrespectiveoftheWBPHresistancetraits.
