Selection of the Mimic Epitopes of Antigens from Schistosoma japonicum by Using Phage Display Techniques

摘要 ToobtainshortpeptidessimulatingantigenicepitopesrelatedtonaturalresistanceagainstSchistosomajaponicum(S.j)inrots,andtoexploretheirimmuneprotectionagainstS.jinmice,phagerandompeptidelibraryof12aminoacidswerescreenedwithpurifiedIgGfromnormalratsere.PositiveclonesthatwereobtainedafterthreeroundsofbioparmingweredetectedbyEI,ISA,andtwoofthemweresequenced.Kunmingmicewereimmunizedwithmixedphageclones.Eachmousewaschallengedwith40±1S.jcercariae,andallmicewereperfused45dayspost-challenge.Thewormsandthelivereggswerecounted.TheresultswerethatthespecificphagesbindingtoIgGwereenriched300foldsafterthreeroundsofbiopanning.TwentyclonesweredetectedbyELISAand19ofthemboundtothespecificIgGofratsera.ThesequenceoftwoclonesrevealednohomologywithothersequencesintheGenBank.Comparedwiththecontrolgroups,thereductionratesofthewormburdenandlivereggwere33.6%and59.8%,respectively.Itwasconcludedthatthespecificpeptides,whichsimulateantigenicmoleculescorrelatedwithnaturalresistancetoS.jinratscouldbeobtainedbyimmunosceeningphagerandompeptidelibraryandaprotectiveimmunityagainstS.jcanbedetectedbytheseepitopesinmice.
