摘要 Objective:Toprovideasoundcellsourceforfurtherex-vivogenetherapyforchronicpain,weattempttodevelopanimmortalizedratastrocytecelllinethatexpressesenkephalinregulatedbydoxycycline.Methods:Retrovirusinfectionmethodwasemployedtodevelopanimmortalizedratastrocytecelllinethatcouldexpressenkephalinregulatedbydoxycycline.ThehPPEgeneexpressionlevelofimmoralizedastroytecells(IAC)/hPPEwasdetectedbyRT-PCR,indirectimmunofluorescencestainingandradioimmunoassay.Results:IACcarryingTet-onsystemtransfectedwithpreproenkephalingenecouldsecreteenkephalinthatwasregulatedbydoxycyclineinadose-dependentmannerandhPPEgeneactivationcouldberepeatedinon-off-oncyclesthroughadministrationorremovalofdoxycycline.Conclusion:Animmortalizedratastrocytecelllinethatsecreteenkephalinunderthecontrolofdoxycyclineisestablishedsuccessfully,whichprovidesaresearchbasisfortransgeniccelltransplantationforanalgesia.
