  • 简介:TherecoveryofGefromlignitebymicroorganismincludestwostages:(1)thebreaking-downofGecomplexofhumusinligniteintosimplecompoundsassistedbymicroorganism;(2)thedesorptionofGecompoundsfromthelignite.TherecoveryrateofGehasbeenenhancedby14%sincethediscoveryofadsorptionanddesorptionofGefromcoal.TheeffectsofpH,leachingagents,andcoalsizeontherecoveryofGewereexperimentallyinvestigated,andtheoptimizedprocessparameterswereobtained.ThereactionheatofGeadsorptionanddesorptioninlignitewasdetermined.Itisabout23-53kJ/mol,whichrevealsthattheadsorptionbelongstophysicalprocess.TherecoveryrateofGefromlignitewithdirectmicroorganismleachingcanreachabout85%,whichishigherthanthatof60%reportedelsewhere.ApotentialprocessforleachingGeinlignitewassuggested.

  • 简介:Thepurposeofthisstudyistodistinguishrespiratorysyncytialvirus(RSV)infectionandimmunologybetweenimmunocompetentandimmunocompromisedmurineandtoexploreimmunemechanismofRSVinfection.AtvarioustimepointsafterRSVinfectionofBALB/cmiceandnudemice,pulmonaryviraltiterswereassayed,RSVantigenwastestedbydirectimmune-fluorescentassayandimmunohistochemistry.PulmonarymRNAexpressionsofTolllikereceptor(TLR)2andTLR4wereassayedbyRT-PCR.CD4+cellsandCD8+cellsinperipheralbloodwereexaminedbyflowcytometryandplasmatotalIgEwasassayedbyELISA.Leukocytesinbronchoalveolarlavagefluid(BALF)andpulmonaryhistologywereidentifiedtoreflectairwayinflammation.ItwasfoundthatRSVtitersofbothmicepeakedonthe3rddaypostinfectionwithamuchhigherlevelofviraltiterinnudemicethaninBALB/cmiceandalongerviraldurationinnudemice(over9dayspostinfection)thaninBALB/cmice(6dayspostinfection).RSVinfectioninducedhigherviralantigenexpressioninnudemice(0.267±0.045)thaninBALB/cmice(0.168±0.031).RSVinfectionenhancedpulmonaryTLR4expressionofBALB/cmice(51.96%±11.34%)andnudemice(48.96%±12.35%)comparedwitheachcontrol(34.04%±10.06%and32.37%±9.87%respectively).CD4+peripheralbloodcellsincreasedinRSVinfectedBALB/cmice(66.51%±2.09%)comparedwiththecontrolBALB/cmice(51.63%±5.90%),andCD4+cellsandCD8+cellsweredeficientinnudemice.RSVinfectionincreasedplasmatotalIgEinbothmice,andBALB/cmicehadalargeramountofIgEonthe7thdaypostinfection(9.02ng/ml±2.90ng/ml)andonthe14thdaypostinfection(12.76ng/ml±4.15ng/ml)thancorrespondingnudemice(3.72ng/ml±1.06ng/mland7.62ng/ml±3.08ng/mlrespectivelyonthe7thand14thdaypostinfection).RSVinfectednudemicehadmoresevereairwayinflammationthaninfectedBALB/cmice.ItisconcludedthatBALB/cmiceandnudemicepresentedsimilarRSVinfectiouscharacteristics.However,

  • 简介:Objective:Infectionofhumanpapillomavirusincondylomaacuminatum(CA)wasdetectedbyrealtimefluorescencequantitativePCR(FQ-PCR)technique.Methods:SpecimensofCA-DNAquantificationfrom94caseswereexaminedbyrealtimeFQ-PCRtechniqueand32caseswerecomparedwiththesamemethodafter10-daystreatment.Results:CA-DNAwasfoundinallpatients,withanaverageof4.0×10^6copies/ul.After10daysoftreatment,theaveragewas2.1×10^5copies/ul.TherewasasignificantdifferenceintheaverageamountofCA-DNAbeforeandafterthetreatment.Conclusion:RealtimeFQ-PCRisagoodmethodforexaminingCA-DNAamountanditcandirectthetreatmentofCA.

  • 简介:Objective:Toexploretheriskfactorsofnosocomialinfectioninseverecraniocerebraltraumaandthewayofprevention.Methods:Theclinicaldataof387patientswithseverecraniocerebraltraumawerereviewed.Results:Thetotalnosocomialinfectionrateofthisstudywas22.99%.Pulmonarynosocomialinfectionpresentedmostfrequently.TheG-bacilliwerethemostcommoninfectiousbacteria.Themortalityrateoftheinfectiongroupwas38.20%.Conclusions:Complicationsofnosocomialinfectionaffecttheprognosisofcraniocerebraltraumapatients.Nosocomialinfectionisrelatedtotheageofthepatients,craniocerebraltraumaseverity,unreasonableutilizationofantibioticsandinvasiveoperations,suchastrachealcannula,mechanicalventilation,urethralcatheterizationanddeepvenouscatheterization.Patientswithseverecraniocerebraltraumashouldbecarefullytreatedandnursedtoavoidnosocomialinfection.Inordertoreducetherateofnosocomialinfection,intensivemeasurementshouldbeadopted.

  • 简介:Objective:Tointroducethecurrentstatusofimplementingsexuallytransmittedinfection(STIs)syndromicmanagementinChina.Methods:Datawerecollectedandanalyzedfromtheliterature.Possiblesolutionsweresuggestedfortheobstaclesencounteredintheimplementationprocess.Results:Validationofrevisedflowchartsforthemanagementofthreesyndromes(urethritis,vaginaldischarge,andgenitalulcers)hasbeenconductedinChina.Thefeasibility,effectivenessandacceptanceofusingasyndromicSTIapproachwereinvestigatedinalargestudyinHainan,ZhejiangandHubeiProvinces.Chiefobstaclestoimplementationincludedthefollowing:lackofsupportfromgovernmentauthorities;negativereactionbyhospitals;lackofcriticalthinkingandinnovation;perceivedover-emphasisontreatmentattheexpenseofprevention;andlackofcoverageforseveralimportantpathogenswithintreatmentalgorithms.Possiblesolutionsmayincludepolicyadvocacy,adoptingsyndromicmanagementintoaChinesecontext,disseminationandpromotion,trainingofproviders,encouraginguseofsyndromicapproach,andconductingoperationalresearch.Conclusions:SyndromicSTImanagementprovidesafeasibleapproachforthediagnosisandtreatmentofSTIsinChina.Furthereffortsshouldbemadeforscalingupitsapplicationinclinicalsettings.

  • 简介:Objectives:ToevaluatetheVidasChlamydia(CHL)assayfordetectingC.Trachomatiswithswabsandfirstcatchurine(FCU)specimensfromSTDpatientsandhighriskpopulations.Methods:Atotalof383patientsweretestedwithtissueculture(TC),VidasCHLandpolymerasechainreaction(PCR)forC.trachomatisonmaleandfemaleswabs,withVidasCHLtestingmaleFCUspecimens.CHLpositiveandequivocalresultswereconfirmedwithablockingassay(CHB).TruepositiveweredefinedaseitherTCpositive,orTCnegtivebutCHLandPCRpositive.TheperformanceofTC,CHLandPCRwereevaluatedaccordingtothisexpandedgoldstandard.Results:Comparedwiththeexpandedgoldstandard,54ofthe232malespecimensweretruepositiveresults.Formaleswabs,TC,CHLandPCRhadsensitivitiesof90.7%,96.3%and94.4%,andspecificitiesof100%,98.3%and97.2%,respectively.Differenceswerenotstatisticallysignificant.FormaleFCUspecimens,CHLsensitivityandspecificitywere83.3%and98.3%;therewaslittledifferencebetweentheseresultsandthatofmatchedswabs.Comparedwiththeexpandedgoldstandard,28ofthe151femaleswabsweretruepositive;TC,CHLandPCRhadsensitivitiesof82.1%,100%and96.4%,andspecificitiesof100%,98.4%and97.6%,respectively.Thedifferencewasalsonotsignificant.Conclusions:VidasCHLassayisverysensitiveandspecificforC.trachomatisdetectionwithswabspecimensofmaleandfemaleSTDpatients.FormaleFCUspecimens,theassayalsohadhighsensitivityandspecificity.CHBmaynotbeneededintheroutinedetectionofChlamydiainfections.PopulationswithhigherincidenceofC.trachomatisinfection.

  • 简介:SincetheoutbreakofaSARSepidemiclastyear,significantadvanceshavebeenmadeonourunderstandingofthemechanismsofinteractionbetweentheSARScoronavirus(CoV)andtheimmunesystem.StronghumoralresponseshavebeenfoundinmostpatientsfollowingSARS-CoVinfection,withhightitersofneutralizingAbspresentintheirconvalescentsera.Thenucleocapsid(N)andspike(S)proteinsofSARS-CoVappeartobethedominantantigensrecognizedbyserumAbs.CD4+TcellresponsesagainsttheNproteinhavebeenobservedinSARSpatientsandanHLA-A2-restrictedcytotoxicTlymphocyteepitopeintheSproteinhasbeenidentified.ItislikelythattheimmuneresponsesinducedbySARS-CoVinfectioncouldalsocausepathologicaldamagetothehost,especiallyinthecaseofproinflammatorycytokines.ThereisalsoevidencesuggestingthatSARS-CoVmightbeabletodirectlyinvadecellsoftheimmunesystem.OurunderstandingontheinteractionbetweenSARS-CoV,theimmunesystemandlocaltissuesisessentialtofuturediagnosis,controlandtreatmentofthisverycontagiousdisease.

  • 简介:ToestablisharapididentificationmethodforcommonpathogenicbacteriaonthebasisofmolecularbiologyandtoconstructapreliminaryPolymeraseChainReaction-CapillaryElectrophoresis-RestrictionFragmentLengthPolymorphism(PCR-CE-RFLP)databaseofbacteriaisolatedfromclinicalspecimensfrequently,183strainscollectedfromclinicalsamplesbelongingto12generaand19specieswhosebiochemicalcharacterizationscorrespondedtothetypicaloneswereexamined.ThegenomicDNAswereamplifiedbytwopairsoffluorescencelabeledprimersaimingat16SrRNAgeneand16S-23SrRNAspacerregiongenerespectivelyatthesametime.PCRproductswerethendigestedbyrestrictionendonucleaseHaeⅢin-completelybeforetakingcapillaryelectrophoresis.TheresultswiththePCR-CE-RFLPpatternsof16SrRNAgeneswerejustalikewithinsomegenera,butwhenitcomesto16S-23SrRNAspacerregiongenes,eachbacteriumshowedauniquepattern,whichcanbedistinguishedfromeachothereasily.ItseemsthatPCR-CE-RFLPpatternsof16SrRNAgenecouldonlybeusedtoclassifythebacteriaintofamilylevel,whereasthedataof16S-23SrRNAspacerregiongenecouldbeutilizedtoidentifythewholemicroorganismsaspreciselyasthespecieslevel.Inspiteofthedataofthespacerregiongenealonecanbesufficientlytoverifythewholebacteria,weinsistthatthe16SrRNAgenecouldbeofsomeassistantincasethatthereshouldbelotsoffamiliesofbacteria,inwhichsomesimilarones,withthesameRFLPdataof16S-23SrRNAspacerregiongene,maycoexist.ThisstudyprovesthattheutilityofPCR-CE-RFLPisaconvenient,rapidmethodtoidentifypathogenicbacteria,andisalsoaquickdiagnosismeasureforapplicationtoclinicaluse.

  • 简介:Object:Toinvestigatetherelationshipbetweenchlamydiatrachomatis(CT)andurogenitalinfection.Method:PositiverateofCTinpatientswithinflammationofurogenitaltractwassignificantlyhigherthanthosewithoutinflammation(P<0.05).Result:Therewasstatisticaldifferenceinthemalesnomattertheywerepatientswithinflammationofurogenitaltractornot(P>0.05),whiletherewasnostatisticaldifferenceinfemales(P>0.05).Theincidenceoftheinfectionwashighamongthoseagingfrom21-50yearsold.Conclusion:TheclinicalmanifestationsofCTinfectionwereobscure,soweshouldexamineCTinpatientswhohavenosymptoms,especiallyinfemalesandthoseofhigh-riskpopulation.
