简介:Aim:TocomparetherecoveryrateofmorphologicallynormalandchromatincondensedspermatozoafromnativesemensamplesusingtheSpermPrepTMfiltrationcolumnsandPercollgradientcentrifugationandtodeterminetheinfluenceofthetwoprocessingtechniquesonfertilizationandpregnancyratesinanIVF-ETprogram.Methods:Sixteensemensamplesobtainedfrompatient'shusbandwereincludedinthisstudy.Eachwasdividedintotwoaliquots.ThefirstaliquotwasprocessedwithSpermPrepTMfiltrationcolumnsandthesecond,Percollgradientcentrifugation.Smearsweremadebeforeandaftersemenprocessingwithbothmethodsfortheevaluationofchromatincondensation(chromomycineCMA3)aswellasmorphology(strictcriteria)ofspermatozoa.Onehundredandseventyoocyteswereretrievedfromthepatientsandtheoocytesfromeachpatientweresubdividedintotwosets:onesetwasinseminatedusingspermatozoaprocessedwithSpermPrepTMandtheotherinseminatedaftersemenprocessingwithPercollgradientcentrifugation.Resuits:ThePercollmethodyieldedasignificantlyhigherpercentageofchromatincondensed(90.8±6.5%vs82.3±8.8%,P=0.017)andmorphologicallynormalspermatozoa(12.9±7.4%vs6.9±4.8%,P=0.001)incomparisontoSpermPrepTM.Whereas,spermcountrecoveryratewassignificantlyhigheraftertheuseofSpermPrepTMthanafterthePercollgradientcentrifugation.Thefertilizationratewassimilarbetweenthetwomethods.Conclusion:SemenprocessingwithPercollshouldberecommendedforintracytoplasmicsperminjectionasthenaturalselectionisbypassedandtheSpermPrepTMtechniquecouldberecommendedfor1VFandIUIprogramsasthespermconcentrationplaysamoresignificantroleintheseprocedures.
简介:Fishmaw(thedriedswimbladdersoffish)isrankedinthelistofthefourseatreasuresinChinesecuisine.Fishmawismainlyproducedfromcroaker,whichisthemosthighlypriced.However,someofthefishmawbeingsoldascroakermawareinfactnotfromcroaker,butfromtheNileperchLatesniloticus.Thepresentworkdeterminedandcomparedtheproximatecomposition,aminoacidandfattyacidcompositionofcroakerProtonibeadiacanthusmawandperchL.niloticusmaw.Theresultsindicatedthatbothmawswerehighproteinsourcesandlowinfatcontent.Thedominantaminoacidsinbothmawswereglycine,proline,glutamicacid,alanineandarginine.Theseaminoacidsconstituted66.2%and66.4%ofthetotalaminoacidsinP.diacanthusandL.niloticus,respectively.TheratioofFAA:TAA(functionalaminoacids:totalaminoacids)inbothmawswere0.69.ThisisagoodexplanationforwhyfishmawshavebeenwidelyutilizedasatraditionaltonicandremedyinAsia.Exceptvalineandhistidine,alltheessentialaminoacidcontentsinP.diacanthuswerehigherthaninL.niloticus.Moreover,croakerP.diacanthusmawcontainedmoreAAandDHAthanperchL.niloticusmaw,showingahigherratioofn-3/n-6,whichismoredesirable.

简介:Inthepresentstudy,weaimedtodeterminethequalityofdifferentbrands(Test1–Test6)ofmefenamicacid,whicharecommerciallyavailableinlocalmarketofKarachi,Pakistan.Variousqualityevaluationtests,includingweightvariation,hardness,thickness,friability,disintegration,assayanddrugrelease,werecarriedout.Resultswerefoundtobeinacceptablelimits.Moreover,releaseprofileswerecomparedusingdifferentdissolutionmedia,suchasphosphatebufferpH6.8,7.4andbiorelevantmedia(FaSSGF,FaSSIF,FeSSIFandSCoF).ReleaseprofilesatpH6.8and7.4wereevaluatedbyone--wayANOVA,model-independentandmodel--dependentmethods.ResultsofANOVAshowedthatnosignificantdifferencewasfoundamongtesterandreferencebrands(Test1–Test6).Similarly,allthebrandswerefoundtobebestfittedwithWeibullmodel.

简介:Therapidincreasingnumberofcompletedbacterialgenomesprovidesagoodopportunitytocomparetheirproteomes.Thisstudywasundertakentospecificallycompareandcontrasttheirsecretomes—thefractionoftheproteomewithpredictedN-terminalsignalsequences,bothtypeIandtypeII.Atotalof176theoreticalbacterialproteomeswereexaminedusingtheExProtprogram.ComparedwiththeGram-positives,theGram-negativebacteriawerefound,onaverage,tocontainalargernumberofpotentialSec-dependentsequences.IntheGram-negativebacteriabutnotintheothers,therewasapositivecorrelationbetweenproteomesizeandsecretomesize,whiletherewasnocorrelationbetweensecretomesizeandpathogenicity.WithintheGram-negativebacteria,intracellularpathogenswerefoundtohavethesmallestsecretomes.However,thesecretomesofcertainbacteriadidnotfitintotheobservedpattern.Specifically,thesecretomeofBorreliaburgdoferihasanunusuallylargenumberofputativelipoproteins,andthesignalpeptidesofmycoplasmasshowclosersequencesimilaritytothoseoftheGramnegativebacteria.Ouranalysisalsosuggeststhatevenforatheoreticalminimalgenomeof300openreadingframes,afractionofthisgenepool(uptoamaximumof20%)maycodeforproteinswithSec-dependentsignalsequences.
