  • 简介:Objective:Toinvestigatetheimmunotherapyefficacyoffusioncells(dendritic-C6anti-TGF-β1cells)inthetreatmentofintracranialgliomas.Methods:Dendriticcellswereisolatedfromratbone-marrowprecursorsstimulatedinvitrowithgranulocyte-macrophagecolony-stimulatingfactor(GM-CSF)andInterleukin-4(IL-4).C6anti-TGF-β1cellsoriginallyfromC6celllineofaratglioblastomaweretransfectedwithplasmidofTGF-β1anti-sensegene.FusionsofdendriticcellsandC6anti-TGF-β1cellswerepreparedbypolyethyleneglycol(PEG).TheDC/C6anti-TGF-β1fusioncellswereobservedandconfirmedbylightmicroscopyandscanningelectronmicroscopy.Experimentalratsweredividedintothreegroupsatrandom:C6cells(I),dendritic-C6anti-TGF-β1fusioncellsandC6cells(II)andIMDMmediumonly(III).Thecellswereinjectedintorightparietalloberegionoftheratwithstereotaxictechnique.Histology,tumornecrosisandsurvivaltimewereevaluated.Results:ComparedwiththeratsthatreceivedC6cells(survivalmediantimewaslessthan20days,tumorregionwasseeninallfieldsofobserved),theratsinjectedwithdendritic-C6anti-TGF-β1fusioncellsandC6cellsgotamoreprolongedlifespan(morethan59days),aswellaslesstumorregion(5.01%-6.2%).Therewasnotumornecrosis,butsomegliaswereseeninsurroundings.Allratsweresurvivedandnonecrosiswasobservedinnegativecontrolgroup.StatisticalanalysisshowedthatgroupIIhadsignificantdifferencecomparedwithgroupI.Conclusions:Dendritic-C6anti-TGF-β1fusioncellscouldprolongthelifespanofrats,providingastrategytoachieveanantitumorresponseagainsttumorsinthecentralnervoussystem.

  • 简介:目的研究BP1C-erbB-2基因在乳腺癌组织中的表达及其相互关系。方法采用RT-PCR方法及免疫组织化学方法检测62例乳腺癌及配对癌旁组织中BP1C-erbB-2基因的表达及雌激素受体(ER)、孕激素受体(PR)的表达。运用x^2检验对各组间差别及相互关系进行分析。结果BP1在乳腺癌和癌旁组织中的阳性表达率分别为64.5%(40/62)和0.05%(3/62),前者明显高于后者,差异具有统计学意义(P〈0.050);BP1阳性表达率在ER阳性乳腺癌组织中为(47.3%)(18/38),在ER阴性乳腺癌组织中为91.7%(22/24),两者之间差异有统计学意义(P〈0.050);BP1阳性表达率在Ⅰ期乳腺癌中为36.3%(4/11),在Ⅱ期乳腺癌中为64.3%(27/42),在Ⅲ期乳腺癌中为100%(9/9),三者间差异有统计学意义(P〈0.050)。在乳腺癌组织中C-erbB-2阳性表达率为46.8%(29/62),C-erbB-2与BP1共阳性者23例,共阴性者16例。结论BP1基因在乳腺癌组织中表达高于癌旁组织,BP1表达与乳腺癌雌激素受体状态及组织学分级有关,与C-erbB-2共表达。

  • 简介:l病例资料患者,女,76岁,因呼吸道疾病入院,常规不规则抗体筛选时3种筛选细胞均呈阳性。妊娠史与输血史:孕3产3活1,头胎正常存活;50年前患者生第2胎时因剖腹产而大量输血,1个月后出现腰背痛、全身不适等反应,新生儿于1周内死亡;生第3胎时新生儿发生严重黄疸,于当天死亡。经过Rh血型系统鉴定、谱细胞检测等判断为IgG抗-D、抗-C混合抗体。

  • 简介:目的探讨蛋白S基因1(proteinS,PROSl)T1378C多态性与新疆地区汉族人群肺血栓栓塞症(Pulmonarythromboembolism,PTE))发生的相关性。方法采用病例-对照研究,病例组为经放射性肺通气/灌注显像和(或)螺旋CT肺动脉造影检查,并结合临床资料确诊的PTE患者100例,对照组为来自同一地区,性别,年龄匹配的健康对照100例。Massarray技术平台进行芯片点制及质谱检测PROS1T1378C的多态性。结果病例组存在TC基因型的频率是8%,对照组TC基因型的频率是1%,差异有统计学意义(χ2=6.703,P〈0.05);病例组C基因型的频率是5%,对照组的等位基因的频率是0.5%,差异有统计学意义(χ2=7.572,P〈0.05)。结论PROS1T1378C多态性与新疆地区汉族人肺栓塞的发生可能有关。

  • 简介:【摘要】目的 探讨3C疗法在儿童1型糖尿病管理中的应用效果。方法将100例于2020年3月-2021年3月期间我院收治的儿童1型糖尿病患儿纳入研究,并以等量电脑随机法均分成A、B两组。A组展开双C疗法治疗,B组展开3C疗法治疗,并对比效果。结果B组患儿血糖改善情况优于A组,血糖达标时间及胰岛素用量均少于A组,对比有统计学意义(P〈0.05)。结论 将3C疗法应用到儿童1型糖尿病患儿的管理中,可助其血糖水平快速达标,并减少日常胰岛素的使用量。

  • 简介:AbstractBackground:NOTCH1 mutation is an essential molecular biologic aberration in chronic lymphocytic leukemia (CLL). CLL patients with NOTCH1 mutation have shown an unfavorable survival and a poor response to chemoimmunotherapy. This study aims to present the mechanisms of adverse prognosis caused by NOTCH1 mutation from the perspective of the splicing factor heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1).Methods:The microarray data in Gene Expression Omnibus datasets were analyzed by bioinformatics and the function of hnRNPA1 was checked by testing the proliferation and apoptosis of CLL-like cell lines. Afterward, quantitative reverse transcription-polymerase chain reaction and Western blotting were applied to explore the relationship among NOTCH1, c-Myc, and hnRNPA1.Results:RNA splicing was found to play a vital part in NOTCH1-mutated CLL cells; hence, hnRNPA1 was selected as the focus of this study. Higher expression of hnRNPA1 validated in primary NOTCH1-mutated CLL samples could promote proliferation and inhibit apoptosis in CLL. The expression of hnRNPA1 increased when NOTCH1 signaling was activated by transfection with NOTCH1 intracellular domain (NICD)-overexpressed adenovirus vector and declined after NOTCH1 signaling was inhibited by NOTCH1-shRNA. Higher expression of c-Myc was observed in NICD-overexpressed cells and hnRNPA1 expression was downregulated after applying c-Myc inhibitor 10058-F4. Moreover, in NICD-overexpressed cells, hnRNPA1 expression decreased through c-Myc inhibition.Conclusion:Overexpression of c-Myc-dependent hnRNPA1 could promote proliferation and inhibit apoptosis in NOTCH1- mutated CLL cells, which might partly account for the poor prognosis of patients with NOTCH1 mutation.

  • 简介:摘要目的对反式转录激活因子Tat第41位赖氨酸(K41)在HIV-1转录活化过程中的作用进行探讨,并对B、C亚型HIV-1中Tat K41的作用进行比较。方法通过荧光素酶活性实验检测野生型Tat、突变型Tat K41A、Tat K41R对HIV-1转录活性的影响,并比较B、C亚型HIV-1中Tat K41作用的差异;通过免疫沉淀(immunoprecipitation, IP)实验检测B、C亚型HIV-1中Tat K41是否影响Tat与SIRT1的结合;通过Western blot检测细胞核内B、C亚型HIV-1的Tat K41是否影响p65 K310的乙酰化水平。结果与B亚型HIV-1不同,C亚型HIV-1的Tat K41突变后显著降低荧光素酶活性,减弱C亚型Tat与SIRT1的结合,降低细胞核内p65 K310ac的水平。结论C亚型Tat K41在HIV-1转录中发挥重要作用。

  • 简介:AbstractBackground:Blood glucose control is closely related to type 2 diabetes mellitus (T2DM) prognosis. This multicenter study aimed to investigate blood glucose control among patients with insulin-treated T2DM in North China and explore the application value of combining an elastic network (EN) with a machine-learning algorithm to predict glycemic control.Methods:Basic information, biochemical indices, and diabetes-related data were collected via questionnaire from 2787 consecutive participants recruited from 27 centers in six cities between January 2016 and December 2017. An EN regression was used to address variable collinearity. Then, three common machine learning algorithms (random forest [RF], support vector machine [SVM], and back propagation artificial neural network [BP-ANN]) were used to simulate and predict blood glucose status. Additionally, a stepwise logistic regression was performed to compare the machine learning models.Results:The well-controlled blood glucose rate was 45.82% in North China. The multivariable analysis found that hypertension history, atherosclerotic cardiovascular disease history, exercise, and total cholesterol were protective factors in glycosylated hemoglobin (HbA1c) control, while central adiposity, family history, T2DM duration, complications, insulin dose, blood pressure, and hypertension were risk factors for elevated HbA1c. Before the dimensional reduction in the EN, the areas under the curve of RF, SVM, and BP were 0.73, 0.61, and 0.70, respectively, while these figures increased to 0.75, 0.72, and 0.72, respectively, after dimensional reduction. Moreover, the EN and machine learning models had higher sensitivity and accuracy than the logistic regression models (the sensitivity and accuracy of logistic were 0.52 and 0.56; RF: 0.79, 0.70; SVM: 0.84, 0.73; BP-ANN: 0.78, 0.73, respectively).Conclusions:More than half of T2DM patients in North China had poor glycemic control and were at a higher risk of developing diabetic complications. The EN and machine learning algorithms are alternative choices, in addition to the traditional logistic model, for building predictive models of blood glucose control in patients with T2DM.

  • 简介:目的探讨血浆或脑脊液中的C-X3-C趋化因子配体1(CX3CL1)是否可作为判断中枢神经系统感染的指标。方法以2015年12月至2016年5月复旦大学附属儿科医院新生儿病房中疑诊中枢神经系统感染行腰椎穿刺检查的新生儿为感染组,根据临床表现,脑脊液常规、生化和培养结果分为中枢感染亚组、败血症亚组和非败血症亚组。以同一时期复旦大学附属妇产科医院产科病房健康新生儿为对照组,根据出生体重分为〈2000、~2500、~3000、~3500和〉3500g亚组,根据孕周分为〈33、~35、~37、~39和〉39周亚组。对照组取新生儿在出生后的脐带血;感染组于急性期或稳定期行腰椎穿刺检查,24h后取静脉血标本。采用Luminex技术检测血或脑脊液标本中的CX3CL1水平,比较各组及其亚组间的差异。结果对照组69例,感染组24例。中枢感染(化脓性脑膜炎)亚组8例,败血症亚组10例,非败血症亚组6例(脑积水2例,泌尿系统感染1例,新生儿惊厥2例、高胆红素血症伴食管气管瘘1例)。对照组脐血CX3CL1水平为(97.8±13.3)pg·mL^-1,脐血CX3CL1水平在不同体重亚组以及不同孕周亚组间差异无统计学意义,P均〉0.05。CX3CL1水平在感染组血浆(95.1±8.2)pg·mL^-1和对照组脐血比较差异无统计学意义(P=0.299)。CX3CL1水平感染组脑脊液中(210.0±11.9)pg·mL^-1高于血浆,差异有统计学意义(P〈0.001)。感染组中的中枢感染亚组、败血症亚组和非败血症亚组脑脊液CX3CL1水平分别为(243.1±13.3)、(208.2±20.1)和(168.7±20.6)pg·mL^-1,3组间差异有统计学意义(P=0.046);中枢感染亚组与非败血症亚组比较,差异有统计学意义(P=0.016);败血症亚组与非败血症亚组比较(P=0.180)、中枢感染亚组与败血症亚组比较(P=0.169),差异均无统计学意义。结论健康新生儿脐带血和感染新生儿外周血CX3CL1表达水平相对稳定

  • 简介:摘要目的探讨hs-CRP(血清超敏C反应蛋白)、HbA1c(全血糖化血红蛋白)和Cys-C(血清胱抑素C)水平变化在糖尿病肾病(DN)早期临床诊断中的意义。方法采用胶乳增强免疫比浊法或酶法在贝克曼AU5800全自动生化分析仪对37例单纯糖尿病患者(单纯DM组)、41例早期DN患者(早期DN组)和40例健康体检者(对照组)的血液样本中的hs-CRP、HbA1c、Cys-C、肌酐(SCr)和尿素(SUrea)检测,并对检测结果进行统计分析。结果单纯DN组和早期DN组检测hs-CRP、HbAlc和Cys-C水平高于对照组,组间比较差异有统计学意义或高度统计学意义(P<0.05或P<0.01);早期DN组检测hs-CRP、HbAlc和Cys-C水平高于单纯DM组,组间比较差异有统计学意义(P<0.05),三组间检测SCr和SUrea水平比较,差异无统计学意义(P>0.05);联合检测的阳性检出率明显高于任意单一指标的阳性检出率,比较差异有统计学意义(P<0.05)。结论hs-CRP、HbAlc和Cys-C可作为DM早期肾功能损伤的重要监测指标,联检有利于提高DN的早期检出率,为临床医生对DN的肾功能损伤早期诊治、病情监测等方面提供有价值的实验室依据。
