  • 简介:ASchwanncellhasregenerativecapabilitiesandisanimportantcellintheperipheralnervoussystem.ThismicroarraystudyispartofabioinformaticsstudythatfocusesmainlyonSchwanncells.Microarraydataprovideinformationondifferencesbetweenmicroarray-basedandexperiment-basedgeneexpressionanalyses.Accordingtomicroarraydata,severalgenesexhibitincreasedexpression(foldchange)buttheyareweaklyexpressedinexperimentalstudies(basedonmorphology,proteinandmRNAlevels).Incontrast,somegenesareweaklyexpressedinmicroarraydataandhighlyexpressedinexperimentalstudies;suchgenesmayrepresentfuturetargetgenesinSchwanncellstudies.Thesestudiesallowustolearnaboutadditionalgenesthatcouldbeusedtoachievetargetedresultsfromexperimentalstudies.Inthecurrentbigdatastudybyretrievingmorethan5000scientificarticlesfromPubMedorNCBI,GoogleScholar,andGoogle,1016(up-anddownregulated)genesweredeterminedtoberelatedtoSchwanncells.However,noexperimentwasperformedinthelaboratory;rather,thepresentstudyispartofabigdataanalysis.OurstudywillcontributetoourunderstandingofSchwanncellbiologybyaidingintheidentificationofgenes.Basedonacomparativeanalysisofallmicroarraydata,weconcludethatthemicroarraycouldbeagoodtoolforpredictingtheexpressionandintensityofdifferentgenesofinterestinactualexperiments.

  • 简介:Themainpathophysiologyofcerebralischemiaisthestructuralalterationintheneurovascularunit,coincidingwithneurovascularmatrixdegradation.Resveratrolhasbeenreportedtobeoneofthemostpotentchemopreventiveagentsthatcaninhibitcellularprocessesassociatedwithischemicstroke.Matrixmetalloproteinases(MMPs)hasbeenconsideredasapotentialdrugtargetforthetreatmentofcerebralischemia.Toexplorethis,wetriedtoinvestigatetheinteractionofresveratrolwithMMPsthroughmoleculardockingstudies.At30minutesbeforeand2hoursaftercerebralischemia/reperfusioninducedbyocclusionofthemiddlecerebralartery,40mg/kgresveratrolwasintraperitoneallyadministered.Afterresveratroladministration,neurologicalfunctionandbrainedemaweresignificantlyalleviated,cerebralinfarctvolumewassignificantlyreduced,andnitriteandmalondialdehydelevelsinthecorticalandstriatalregionsweresignificantlydecreased.ThemoleculardockingstudyofresveratrolandMMPsrevealedthatresveratroloccupiedtheactivesiteofMMP-2andMMP-9.Thebindingenergyofthecomplexeswas–37.848672kJ/moland–36.6345kJ/molforMMP-2andMMP-9,respectively.IncaseofMMP-2,Leu164,Ala165andThr227wereengagedinH-BondingwithresveratrolandincaseofMMP-9,H-bondingwasfoundwithGlu402,Ala417andArg424residues.ThesefindingscollectivelyrevealthatresveratrolexhibitsneuroprotectiveeffectsoncerebralischemiathroughinhibitingMMP-2andMMP-9activity.

  • 简介:Acupunctureiswidelyusedtotreatfunctionaldyspepsiawithsatisfactoryoutcomes.CombinationoftheHeandMuacupointsiscommonlyusedandhasasynergisticeffectonfunctionaldyspepsia;however,itsunderlyingmechanismsremainunclear.Therefore,arandomizedcontrolledparallelclinicaltrialiscurrentlyunderwayatChengduUniversityofTraditionalChineseMedicine,China.ThistrialisdesignedtoexploretheefficacyofandcentralresponsestotheHe-Mupointcombinationinpatientswithfunctionaldyspepsiausingfunctionalmagneticresonanceimaging.Atotalof105patientswithfunctionaldyspepsiawillbeallocatedinto3groups:thelow-Hepointgroup(puncturingatZusanli(ST36)),Mupointgroup(puncturingatZhongwan(CV12)),andHe-Mupointcombinationgroup(puncturingatST36andCV12).Everyparticipantwillreceive20sessionsofmanualacupuncturefor4weeks.Theneedleswillbeinsertedperpendicularlytoadepthof1to2cun.Theangleofrotationandtwistingwillrangefrom90to180degrees,whileliftingandthrustingwillrangefrom0.3to0.5cm.Thevariousmanipulationswillbeperformed60to90timesperminute.Theneedleswillremaininplacefor30minutes,duringwhichmanipulationwillbeappliedevery10minutes.Magneticresonanceimagingwillbeperformedbeforeandafter20sessionsofacupuncture.TheprimaryoutcomeissymptomimprovementaccordingtotheChineseversionoftheNepeanDyspepsiaIndex.SecondaryoutcomesincludetheLeedsdyspepsiaquestionnaire,Self-RatingAnxietyScale,Self-RatingDepressionScale,BeckAnxietyInventory,BeckDepressionInventory,andvisualanaloguescalescoresbeforeandafter10and20sessionsofacupuncture.Needlesensationandadverseeventswillbeusedtoassessthetherapeuticeffects.ThisstudywillpromotemorewidespreadawarenessofthebenefitsofacupointcombinationintheclinicalsettingandprovideafurtherexplanationoftheneuromechanismbywhichacupunctureattheHe-Mupointcombinationforfunctio

  • 简介:Abducensnervepalsy(ANP)iscommonlyseeninpatientswithdiabetesmellitus.ThevalidityofacupunctureasatraditionalChinesemedicinemethodinperipheralnerverepairiswellestablished.However,itsefficacyinrandomizedcontrolledtrialsremainsunclear.Herein,wedesignedaprotocolforaprospective,single-center,randomizedcontrolledtrialtoinvestigatetheeffectofintraorbitalelectroacupunctureondiabeticANP.Weplantorecruit60patientswithdiabeticANP,andrandomlydividethemintotreatmentandcontrolgroups.Patientsinbothgroupswillcontinuetheirglucose-loweringtherapy.Aneuralnutritiondrugwillbegiventobothgroupsforsixweeks.Thetreatmentgroupwillalsoreceiveintraorbitalelectroacupuncturetherapy.Wewillassessefficacyoftreatment,eyeballmovement,diplopiadeviationandthelevelsoffastingblood-glucoseandglycosylatedhemoglobinbeforetreatmentat2,4,and6weeksaftertreatment.Theefficacyandrecurrencewillbeinvestigatedduringfollow-up(1monthafterintervention).ThisprotocolwasregisteredatChineseClinicalTrialRegistryon16January2015(ChiCTR-IPR-15005836).ThisstudywasapprovedbytheEthicsCommitteeofFirstAffiliatedHospitalofHarbinMedicalUniversityofChina(approvalnumber:201452).AllprotocolswillbeinaccordancewithDeclarationofHelsinki,formulatedbytheWorldMedicalAssociation.Writteninformedconsentwillbeprovidedbyparticipants.WeenvisagethattheresultsofthisclinicaltrialwillprovideevidenceforpromotingclinicaluseofthisnewtherapyformanagementofANP.

  • 简介:BACKGROUND:ItisknownthatacupuncturetherapycandecreaseplasmaneuropeptideY(NPY)levelsinpatientswithcerebralinfarction,butdifferenttypesofacupuncturetherapyusedinvariousstagesofcerebralinfarctionhavenotbeenevaluated.OBJECTIVE:Toexploretheeffectofacupuncturetherapyonresuscitation(XingnaoKaiqiao)andplasmaNPYlevelsinpatientswithveryearlystageacutecerebralinfarction.DESIGN,TIMEANDSETTING:Thiscase-controlledstudywasperformedattheAffiliatedHospitaloftheMedicalCollegeoftheChinesePeople'sArmedPoliceForcebetweenSeptember2004andOctober2005.PARTICIPANTS:Sixtypatientswithacutecerebralinfarctionof≤6hourswereusedinthisstudy.Patientswererandomlydividedintoanacupuncturetherapygroup(n=30)andaroutinetreatmentgroup(n=30).Another30healthysubjectswereusedasthecontrolgroup.METHODS:TheacupuncturetherapyofXingnaoKaiqiaousedintheacupuncturetherapygroupwasbasedonroutinewesternmedicaltreatmentandwasperformedatbilateralNeiguan(PC6)usingthetwirling,reinforcing-reducingmethod,Renzhong(DU26)usingheavybird-peckingneedling,Sanyinjiao(SP6)usingreinforcingandreducingbyliftingandthrustingtheneedle,Jiquan(HT1),Weizhong(BL40)andChize(LU5)usingreinforcingandreducingbyliftingandthrustingtheneedle.Theacupuncturelastedfor14days.Patientsintheroutinetreatmentgroupunderwentroutinemedicaltreatmentandnointerventionwasgiventosubjectsinthecontrolgroup.MAINOUTCOMEMEASURES:A4mLvenousbloodsamplewasobtainedatdifferenttimepoints,i.e.,immediatelyafterhospitalization,thenextmorning,7and14daysaftertreatment,tomeasureplasmaNPYlevelspre-andpost-treatmentusingtheradio-immunitymethod.RESULTS:TheplasmaNPYlevelsweresignificantlyhigherinboththeroutinetreatmentgroupandtheacupuncturetherapygroupthaninthecontrolgrouppre-andpost-treatment(P<0.01).Inparticular,theplasmaNPYlevelsinboththeacupuncturetherapygroupa

  • 简介:Moststudiesaddressingthespecificityofmeridiansandacupuncturepointshavefocusedmainlyonthedifferentneuraleffectsofacupunctureatdifferentpointsinhealthyindividuals.Thisstudyexaminedtheeffectsofacupunctureonbrainfunctioninapathologicalcontext.Sixteenpatientswithischemicstrokewererandomlyassignedtotruepointgroup(trueacupunctureatrightWaiguan(SJ5))andshampointgroup(shamacupuncture).Resultsoffunctionalmagneticresonanceimagingrevealedactivationinrightparietallobe(Brodmannareas7and19),therighttemporallobe(Brodmannarea39),therightlimbiclobe(Brodmannarea23)andbilateraloccipitallobes(Brodmannarea18).Furthermore,inhibitionofbilateralfrontallobes(Brodmannarea4,6,and45),rightparietallobe(Brodmannareas1and5)andlefttemporallobe(Brodmannarea21)wereobservedinthetruepointgroup.Activationintheprecuneusofrightparietallobe(Brodmannarea7)andinhibitionoftheleftsuperiorfrontalgyrus(Brodmannarea10)wasobservedintheshamgroup.Comparedwithshamacupuncture,acupunctureatWaiguaninstrokepatientsinhibitedBrodmannarea5onthehealthyside.Resultsindicatedthatthealteredspecificityofsensation-associatedcortex(Brodmannarea5)ispossiblyassociatedwithacentralmechanismofacupunctureatWaiguanforstrokepatients.
