  • 简介:PolysaccharidesfromnumeroustraditionalChinesemedicineshavebeenprovenasthebioactiveingredientsandarehenceusedasthequalitycontrolmarkers.However,theassessmentcriteriaalwaysshowapoorspecificity,duetothelackofmaticcomparisonamongtheanalogousherbs.Inthepresentstudy,twosimilarmaterials,namelysea-tangleandsargassum,wereselectedasthemodelherbstodevelopmorespecificmethodsforqualitycontrol.Twowell-establishedmethods,determinationofthetotalpolysaccharidescontentandmonosaccharidescompositionanalysis,werebothemployed.Baseduponthequantitativeresults,theevaluationcriteriaofthepolysaccharidescontentsofnotlessthan2.0%and1.7%wereproposedforsea-tangleandsargassum,tively.NineidenticalmonosaccharidederivativesappearedontheHPLCchromatogramsofthehydrolysisandderivatizedsolutionsofthetwodrugs.Principalcomponentanalysisandorthogonalpartialleastsquaresdiscriminantanalysisusingthepeakareasofmonosaccharidesderivativesasthevariableswereperformed,andtheresultsindicatedthatmannuronicacidandxylosewiththeoppositeconcentrationsinthetwodrugswerethedifferentialcomponents.Adiscriminativecriterionusingthepeakarearatioofthesetwomonosaccharidesderivativeswasproposedforthequalitativeidentification.Inconclusion,amorespecificandquantitativequalitycontrolmethodwasdevelopedforsea-tangleandsargassum.

  • 简介:PhyllanthusUrinariaL.(PUL)isatraditionalChinesemedicineusedtotreathepaticandrenaldisorders.However,themechanismofitshepatoprotectiveactionisnotfullyunderstood.Inthepresentstudy,bloodbiochemicalindexesandliverhistopathologicalchangeswereusedtoestimatetheextentofhepaticinjury.GC/MSandLC/MS-baseduntargetedmetabolomicswereusedincombinationtocharacterizethepotentialbiomarkersassociatedwiththeprotectiveactivityofPULagainstCCl_4-inducedliverinjuryinrats.PULtreatmentcouldreversetheincreaseinALT,ASTandALPinducedbyCCl_4andattenuatethepathologicalchangesinratliver.SignificantchangesinlivermetabolicprofilingwereobservedinPUL-treatedgroupcomparedwithliverinjurymodelgroup.SeventeenbiomarkersrelatedtothehepatoprotectiveeffectsofPULagainstCCl_4-inducedliverinjurywerescreenedoutusingnonparametrictestandPearson'scorrelationanalysis(OPLS-DA).TheresultssuggestedthatthepotentialhepatoprotectiveeffectsofPULinattenuatingCCl_4-inducedhepatotoxicitycouldbepartiallyattributedtoregulatingL-carnitine,taurocholicacid,andaminoacidsmetabolism,whichmaybecomepromisingtargetsfortreatmentoflivertoxicity.Inconclusion,thisstudyprovidesnewinsightsintothemechanismofthehepatoprotectionofPhyllanthusUrinaria.

  • 简介:SophoraeFlavescentisRadix(SophoraflavescensAit.,SFR)andSophoraeTonkinensisRadixetRhizoma(S.tonkinensisGapnep.,STR)aretwocommonlyusedtraditionalChinesemedicinesfromSophora(Leguminosae)plants,whicharebelievedtopossesssimilarbioactivecomponentswithentirelydifferentclinicalapplications.InordertofindoutthecharacteristicchemicalconstituentspotentiallyleadingtotheuniquemedicinalpropertiesclaimedforeachofthetwocloselyrelatedTCMs,anHPLCfingerprintmethodwasdevelopedforanalysesofthealkaloidandflavonoidconstituentsofSFRandSTR,respectively,whichwerefurtherevaluatedandcomparedthroughsimilaritycalculationandhierarchicalclusteringanalysis(HCA).Theresultsfromthepresentstudyshowedthatthealkaloidfingerprintsofthetwoherbsweresimilar,withmanycomponentsco-existinginbothdrugsandvariousbatchesofsamplesfromdifferentspeciesbeingmixedtogetherintheHCAdendrogram.However,theirflavonoidconstituentsweretotallydifferentwithspecificfingerprintsbeingyieldedforeachherb,andfurtherHCAanalysisshowedthatthetestedsamplescouldalmostbeclearlydividedintotwogroupsbasedontheiroriginsofspecies.TheresultsfromthepresentstudyindicatedthattheflavonoidconstituentscouldserveasthedifferentiallydiagnosticconstituentsofSFRandSTRandmightpotentiallyattributedtotheirdistincttherapeuticeffects.
