  • 简介:Objective:Estimationofactivityaccumulatedintumorandorgansisveryimportantinpredictingtheresponseofradiopharmaceuticalstreatment.Inthisstudy,wesynthesized177Lutetium(177Lu)-trastuzumabironoxidenanoparticlesasadoubleradiopharmaceuticalagentfortreatmentandbetterestimationoforganactivityinanewwaybymagneticresonanceimaging(MRI).Methods:177Lu-trastuzumab-ironoxidenanoparticlesweresynthesizedandallthequalitycontroltestssuchaslabelingyield,nanoparticlesizedetermination,stabilityinbufferandbloodserumupto4d,immunoreactivityandbiodistributioninnormalmiceweredetermined.Inmicebearingbreasttumor,liverandtumoractivitieswerecalculatedwiththreemethods:singlephotonemissioncomputedtomography(SPECT),MRIandorganextraction,whichwerecomparedwitheachother.Results:Thegoodresultsofqualitycontroltests(labelingyield:61%±2%,meannanoparticlehydrodynamicsize:41±15nm,stabilityinbuffer:86%±5%,stabilityinbloodserum:80%±3%,immunoreactivity:80%±2%)indicatedthat177Lu-trastuzumab-ironoxidenanoparticlescouldbeusedasadoubleradiopharmaceuticalagentinmicebearingtumor.Resultsshowedthat177Lu-trastuzumab-ironoxidenanoparticleswithMRIhadtheabilitytomeasureorganactivitiesmoreaccuratethanSPECT.Conclusions:Co-conjugatingradiopharmaceuticaltoMRIcontrastagentssuchasironoxidenanoparticlesmaybeagoodwayforbetterdosimetryinnuclearmedicinetreatment.

  • 简介:Inthepresentanalysisontherelationshipsamongthedepthoflithospherebrittlefracture,seismotectonicsandgeothermalanomalousactiveinTibetanplateauwereinvestigatedusingtheseismicdadafromISCandChineseseismicnetandgeothermaldata.Theresultssuggestthattheregionofanomalouslygeothermalactivityalmostcoincideswiththatofthenormalfaultingtypeearthquake.Thegeothermalanomalyactivityregioncoincidesspatiallywiththatoftheeventsdeeperthan60kmaswellas.Thenormalfaultingearthquakesmaybemainlytectonicactivityregimesuntil110kmdeepinthethermalanomalyregion.ThestrikedirectionsofeventsarelikelytheN-Sdirection,coincidingwiththestrikeofthethermalanomalyactivebelts.Theearthquakesalignalongthenormalfaultsandfaulted-depressionzonewiththeN-Sdirection.Thethermalanomalyactivityalsodistributesalongthefaulted-depressionzone.Manyeventsdeeperthan60kmexistintheanomalouslygeothermalactivityregionintheplateau.Eventsextendtobottomofthelithosphereof110kmfromthesurface,likecolumnarseismiccrowd.ThelithosphereextendsalongtheE-WdirectionduetotheE-WextensionalstressinthecentralandsouthernTibetanplateau,altitudeoftheplateau.ThetensionalstressintheE-WresultsinthelithospherefracturesandthenormalfaultsstrikingN-Sdirection,grabensandfaulted-depressionzones.Thermalmaterialfromtheasthenospherewellsupwardtothesurfacealongdeepseismicfracturesandfaultsthroughthethickcrust.TheanomalouslythermalactivitiesareattributabletotheupwellingthermalmaterialfromthemantleinthealtitudeofTibetanplateau.

  • 简介:ByapplyinganewvortexdetectionmethodtotheECMWF40-yrreanalysis(ERA40)datafrom1985to2002,theclimatologyofsummervorticeshasbeeninvestigatedinfivesubtropicalregions,i.e.,thenorthwesternPacific,northeasternPacific,northwesternAtlantic,northeasternAtlantic,andAustralia-SouthPacific,followedbyvalidationwithNCEP/NCARreanalysisdata.Resultsareasfollows:(1)ThespatialdistributionsofERA40vortexactivities(VAC)werewellconsistentwiththoseofNCEP/NCARreanalysis(NRA)resultsinallregions,especiallyinnorthwesternPacific.(2)Becauseofdifferentmodelresolutions,boththenumberandintensityofvorticesobtainedfromNRAweresignificantlyweakerthanERA40's.(3)Vorticesmainlycruisedincoastsandtheadjacentseas,fromwheretothelandortheopenseavortexactivitiesweregraduallydecreased.(4)ThereweretwoactivecentersinthenorthwesternPacific:onewaslocatedinSouthChinaSeaandtheother,asthelargestcenterofthefiveregions,spreadfromtheeastsideofthePhilippinestoJapan.(5)OverthenorthwesternAtlantic,mostvorticesoccurredinPanamaanditswest-sideoffshore.(6)ThespatialdistributionsofvorticeswerealikebetweenthenortheasternPacificandnortheasternAtlantic,bothspreadingfromcoaststothewest-sideseaat5°-20°N.(7)IntheAustralia-SouthPacific,vorticeswerenotasactiveasthoseintheotherfourregions,andmostlytookplaceintheequator-sideofnearoceanareas.(8)ExceptthenorthwesternPacificandnorthwesternAtlantic,theVACinterannualvariationsintheotherthreeregionsweredifferentbetweenERA40andNRAdata.(9)InthenorthwesternPacificandnorthwesternAtlantic,theVACinterannualvariationcouldbeseparatedtoseveraldistinctstages.(10)Sincethemid1980s,meanvortexintensitywasgettingincreasedinthenorthwesternPacific,whichwasmostsignificantinthesubtropicalareasonaglobalbasis.InthewesternNorthAtlantic,therewasadecreasing(increasing)trendofthemeanvortexintensitybefo

  • 简介:EuphorbiakansuiisacommonlyusedtraditionalChinesemedicineforthetreatmentofedema,pleuraleffusion,andasthma,etc.Accordingtothepreviousresearches,terpenoidsinE.kansuipossessvariousbiologicalactivities,e.g.,anti-virus,anti-allergy,antitumoreffects.Inthiswork,twentyfiveterpenoidswereisolatedfromE.kansui,includingthirteeningenane-andeightjatrophane-typediterpenoids(withtwonewcompounds,kansuininPandQ)andfourtriterpenoids.EighteenofthemwereanalyzedbyMTSassayforinvitroanticanceractivityinfivehumancancercelllines.Structure-activityrelationshipfor12ingenane-typediterpenoidsincolorectalcancerColo205cellswerepreliminarystudied.Significantanti-proliferationactivitieswereobservedinhumanmelanomacellsbreastcancerMDA-MB-435cellsandColo205cells.Morethanhalfoftheisolatedingenane-typediterpenoidsshowedinhibitoryactivitiesinMDA-MB-435cells.Eightingenane-andonejatrophane-typediterpenoidspossessedmuchlowerIC_(50)valuesinMDA-MB-435cellsthanpositivecontrolstaurosporine.Preliminarystructure-activityrelationshipanalysisshowedthatsubstituentonposition20wasimportantfortheactivityofingenane-typediterpenoidsinColo205cellsandsubstituentonposition3contributedmoresignificantbiologicalactivityofthecompoundsthanthatonposition5inbothMDA-MB-435andColo205cells.

  • 简介:Zygosaccharomycesrouxiiisasalt-tolerantyeastspeciescapableofremovingcadmium(Cd)pollutantfromaqueoussolution.Presently,thephysiologicalcharacteristicsofZ.rouxiiunderthestressofsodiumchloride(NaCl)andCdarepoorlyunderstood.ThisstudyinvestigatedtheeffectsofNaClandCdonthegrowth,oxidativestressandantioxidantenzymeactivitiesofZ.rouxiiafterstresstreatmentfor24h.ResultsshowedthatNaClorCdalonenegativelyaffectedthegrowthofZ.rouxii,butthegrowth-inhibitingeffectofCdonZ.rouxiiwasreducedinthepresenceofNaCl.FlowcytometryassayshowedthatunderCdstress,NaClsignificantlyreducedtheproductionofreactiveoxygenspecies(ROS)andcelldeathofZ.rouxiicomparedwiththoseintheabsenceofNaCl.Theactivitiesofsuperoxidedismutase(SOD),catalase(CAT),andperoxidase(POD)ofZ.rouxiiweresignificantlyenhancedby2%–6%NaCl,whichlikelycontributedtothehighsalttoleranceofZ.rouxii.ThePODactivitywasinhibitedby20mgL-1CdwhiletheSODandCATactivitieswereenhancedby8mgL-1Cdandinhibitedby20mgL-1or50mgL-1Cd.Theinhibitoryeffectofhigh-levelCdontheantioxidantenzymeactivitiesofZ.rouxiiwascounteractedbythecombineduseofNaCl,especiallyat6%.ThisprobablyaccountedforthedecreaseinCd-inducedROSproductionandcelldeathofZ.rouxiiafterincubationwithNaClandCd.OurworkprovidedphysiologicalcluesastotheuseofZ.rouxiiasabiosorbentforCdremovalfromseawaterandliquidhighlysaltyfood.

  • 简介:Theeffectsofpolishingonproximatecompositions,physico-chemicalcharacteristics,mineralcompositionsandantioxidantpropertiesofthericefloursobtainedfromthreedifferentpigmentedricevarieties(Chak-haoAngangba,Chak-haoAmubiandChak-haoPoireiton)wereinvestigated.Thericevarietiesweresignificantly(P<0.05)differentinthecontentsofthetestcharacteristics.Lipids,ash,minerals,phytochemicals(phenolicacidsandflavonoids)and2,2-diphenyl-1-picrylhydrazyl(DPPH)activityofricefloursweredecreasedafterpolishing(9%degreeofmilling),whileamylosecontentandlightnesswereincreased.X-raydiffractionpatternofricefloursexhibitedA-typecrystallinepatternwithreflectionsat15.1o,17.1o,18.2oand23.0o.Pastingpropertiesandtransitiontemperaturesweredecreasedafterpolishingtreatment.Polishingresultedinchangesinthecrystallinity,enthalpyandmorphologyofriceflours.
