  • 简介:BasedonAVISO(archiving,validationandinterpretationofsatellitedatainoceanography)datafrom1993to2010,QuikSCAT(QuickScatterometer)datafrom2000to2008,andArgodatafrom2003to2008,theinterannualvariabilityoftheGreatWhirl(GW)andrelatedmechanismsarestudied.ItshowsthattheoriginandterminationtimesoftheGW,aswellasitslocationandintensity,havesignificantinterannualvariability.TheGWappearedearliest(latest)in2004(2008)andvanishedearliest(latest)in2006(2001),withtheshortest(longest)durationin2008(2001).Itscenterwasmostsouthward(northward)in2007(1995),whiletheminimum(maximum)amplitudeandareaoccurredin2003and2002(1997and2007),respectively.TheGWwasweakeranddisappearedearlierwithitslocationtendingtobeinthesouthwestin2003,whilein2005itwasstronger,vanishedlaterandtendedtobeinnortheast.TheabnormalyearswereoftennotthesameamongdifferentcharactersoftheGW,andwerenotallcoincidentwithENSO(ElNi?o-SouthernOscillation)orIOD(IndianOceanDipole)events,indicatingtheverycomplexnatureofGWvariations.MechanisminvestigationsshowsthattheinterannualvariabilityofintraseasonalwindstresscurlinGWregionresultsinthatoftheGW.ThegenerationoftheGWiscoincidentwiththearrivalofRossbywavesattheSomalicoastinspring;theintensityoftheGWisalsoinfluencedbyRossbywaves.TheterminationoftheGWcorrespondswelltothesecondoneofthetoptwopeaksinthebaroclinicenergyconversionrateinGWregion,andtheintensityandthepositionoftheGWarealsocloselyrelatedtothetoptwobaroclinicenergyconversionrates.

  • 简介:Inthisstudy,weexaminedtheeffectofelevatedtemperatureontheexpressionpatternsofgenes,i.e.,nacrein,irr,n16,n19,andhsp70inthepearloysterPinctadafucata.Theexperimentwascarriedoutat4temperatures,i.e.,20℃(ambient,control),24,28℃,and32℃.TheexpressionlevelsoftargetgenesinP.fucatawereassayedat0,6,24,48,and96hviareal-timepolymerasechainreaction.Resultsshowedthattheexpressionlevelsofnacreinandirrhadnosignificantvariationsamongdifferenttimepointsbelow28℃,butsignificantlyincreasedovertimeat32℃.Theexpressionlevelsofn16andn19didnotchangemarkedlyat20℃.Theformerincreasedsignificantlyat6hand24hwhilethelattersubstantiallydecreasedduring6–96hat24,28and32℃.Amongdifferenttemperatures,thelevelofn16wassignificantlylowerat20℃thanatothertemperaturesduring6–96h,andthelevelofn19significantlyvariedamongdifferenttemperaturesat48hand96h.Theexpressionlevelofhsp70wassignificantlyhigherat32℃thanat20,24and28℃at24h.TheseresultsdemonstratedthatelevatedtemperatureimpactedthephysiologicalprocessesofP.fucataandpotentiallyinfluenceditsadaptabilitytothermalstress.

  • 简介:EvolutionaryevidencesuggeststhatSox3,amemberofthehigh-mobility-group(HMG)familyoftranscriptionfactors,isanancestralprecursorofSryandisinvolvedinsexdeterminationsimilartoSry.However,thereislimitedinformationregardingtheSOX3geneoftheblackrockfish(Sebastesschlegeli).Inthisstudy,wefirstisolatedSOX3genefromthegonadsofS.schlegelibyhomologycloning.Thefull-lengthofS.schlegeliSOX3(SsSOX3)cDNAwas1386bp,comprisinga906-bpopenreadingframe,whichencodesapeptideshowing93.6%and93.9%homologywiththeSox3proteinsofEpinepheluscoioidesandOryziaslatipe,respectively.ComparisonofthecDNAsequenceoftheSsSOX3genewiththecorrespondinggenomicDNAfragmentrevealedthattheSsSOX3geneconsistsofasingleexon.PhylogeneticanalysisdemonstratedtheevolutionaryrelationshipofSsSOX3withotherknownSOXB1genesinfishandtetrapods.ThepromoterregioncontainsbindingsitesofseveraltranscriptionalfactorsthatmightparticipateintheregulationofSsSOX3expression.Quantitativereal-timePCRanalysisindicatedthatSsSOX3wasexpressedinalltheinvestigatedlarvaldevelopmentalstagesfrom1to35daysafterbirthandthelevelofexpressiongraduallydecreasedasthedevelopmentproceeded.SsSOX3exhibitedsexuallydimorphicexpressioninadultgonads,withhighexpressionintheovarybutlowexpressioninthetestis.InsituhybridizationrevealedthatSsSOX3wasstronglyexpressedinoocytesandfollicularcellsofovariesbutslightlyexpressedingermcellsoftesticulartissues.Therefore,thisstudysuggeststhatSsSOX3playsanimportantroleinoogenesisandovarydifferentiationinS.schlegeli.

  • 简介:ThisstudyexaminedtheeffectofsalinityontheexpressionofNa+/K+-ATPase(NKA)α-subunitandvacuolar-typeH+-ATPase(V-ATPase)β-subunitgeneinthegillofLitopenaeusvannamei.Semi-quantitativereversetranscription-polymerasechainreaction(RT-PCR)assayshowedthattheexpressionofNKAα-subunitandV-ATPaseβ-subunitgenewassignificantlyinfluencedbysalinity.ItwasfoundthattheNKAactivitysignificantlyvariedwithsalinityintimeanddosedependentmanner;whereastheV-ATPaseactivitydidnot.TheabundanceofNKAα-subunitgenetranscriptincreasedrapidlywhenthesalinitydecreasedfrom26bto21,andslowlywhenthesalinitydecreasedfrom26to31withinthefirst24h.Whenthesalinitydecreasedfrom26to21,thetranscriptionofNKAα-subunitgeneingillepitheliumwashigherat12hthanthatat0h,whichwasconsistentwiththeresultofimmunoblottingassayofNKAα-subunit.Inaddition,salinityhadasignificanttime-anddose-dependenteffectontheconcentrationofbiogenicaminesinbothhemolymphandgill.Ascomparedtootherparameters,theconcentrationofdopamine(DA)and5-hydroxytryptamine(5-HT)variedindifferentpatternswhenthesalinitydecreasedfrom26to21orincreasedfrom26to31,suggestingthatDAand5-HTplayeddifferentregulatoryrolesinosmoticadaptionandmodulationofshrimpwhensalinityvaries.
