  • 简介:AbstractInfectious diseases are an increasing threat to global biosafety. Vaccination is the most effective and cost-efficient method for preventing and controlling infectious diseases. The development of new vaccines is inextricably linked to the advancement of materials that serve as essential components of vaccines, such as antigens, adjuvants, and their carriers. The physicochemical and biological properties of vaccines—such as the kinetics of antigen retention and presentation—are determined by the material compositions of vaccines and carriers, affecting the overall efficacy. The sustained release of antigens prolongs their retention time in germinal centers and improves humoral immune responses. Pulsatile release that imitates clinical dosing regimens can improve patient adherence to vaccination, affording increased vaccine coverage. Herein, we review progress of materials innovation on altering vaccine release kinetics, which affects the overall vaccine efficacy, safety, and compliance.

  • 简介:AbstractAnti-influenza drugs are one of the most critical pathways for control of influenza virus infection. Drugs that have been developed or are developing may function via different mechanisms, and so far, inhibitors of influenza virus polymerase are among the most promising types of drugs. Favipiravir and Baloxavir, also named T-705 and Xofluza respectively, have been approved for influenza treatment in Japan and the United States. Favipiravir effectively and selectively inhibits the RNA-dependent RNA polymerase (RdRp) of RNA viruses while Baloxavir specifically targets the cap-dependent endonuclease PA of influenza viruses. These two drugs have been suggested as the first candidate drugs for influenza infection treatment, especially for strains resistant to other anti-influenza drugs. This review will focus on the pharmaceutical mechanisms and anti-influenza activity of these two drugs.

  • 简介:Baseduponstudiesfromrandomizedclinicaltrials,theextended(D2)lymphnodedissectionisnowrecommendedasastandardprocedureforlocaladvancedgastriccancerworldwide.However,therationalextentlymphadenectomyforlocaladvancedgastriccancerhasremainedatopicofdebateinthepastdecades.Duetothelimitationoflowmetastaticrateinpara-aorticnodes(PAN)inJCOG9501,theclinicalbenefitofD2+para-aorticnodaldissection(PAND)forpatientswithstageT4and/orstageN3disease,whichisverycommoninChinaandothercountriesexceptJapanandKorea,cannotbedetermined.Furthermore,theroleofsplenectomyforcompleteresectionofNo.10andNo.11nodeshasbeencontroversial,andhowever,thefinalresultsfromtherandomizedtrialofJCOG0110haveyettobecompleted.GastriccancerwiththeNo.14andNo.13lymphnodemetastasisisdefinedasM1stageinthecurrentversionoftheJapaneseclassification.WeproposethatD2+No.14vand+No.13lymphadenectomymaybeanoptioninapotentiallycurativegastrectomyfortumorswithapparentmetastasistotheNo.6nodesorinfiltratetoduodenum.Theexaminedlymphnodeandextranodalmetastasisaresignificantlyassociatedwiththesurvivalofgastriccancerpatients.

  • 简介:AbstractGastric cancer, which has a high incidence and poor prognosis, remains a therapeutic challenge. Recently, neoadjuvant therapy has attracted increasing attention due to high recurrence rate and low survival rate after resection in most patients with advanced stage. Clinical trials show that neoadjuvant approaches confer a significant survival advantage for resectable locally advanced gastric cancer. The specific advantages of chemoradiotherapy compared with chemotherapy have not been clarified; optimal regimens and cycles, particularly in the preoperative setting, should be studied further; and trials aimed at determining the role of targeted and immunological therapies should be conducted.

  • 简介:Theuterinetetaniccontractionanduterinearterybloodflowreductionarepossiblereasonsforprimarydysmenorrhea(PD).Inthepresentstudy,weaimedtoevaluatetheuterinerelaxanteffectandtheinfluenceonuterinearterybloodvelocityofGe-GenDecoction(GGD),awell-knownChineseherbalformula.InfemaleICRmice,uterinecontractionwasinducedbyoxytocinexposurefollowingestradiolbenzoatepretreatment,andtheuterinearterybloodvelocitywasdetectedbyDopplerultrasound.HistopathologicalexaminationoftheuterinetissuesampleswereperformedbyH&Estaining.Exvivostudiesdemonstratedthatoxytocin,posteriorpituitary,oracetylcholineinducedcontractionsinisolatedmouseuterus.GGDinhibitedbothspontaneousandstimulatedcontractions.InvivostudydemonstratedthatGGDsignificantlyreducedoxytocin-inducedwrithingresponseswithamaximalinhibitionof87%.FurtherstudydemonstratedthatGGDnormalizedoxytocin-inducedabnormalitiesofprostaglandinsF2alpha(PGF2α)andCa2+inmice.Inaddition,injectionofoxytocininducedadecreaseinuterinearterybloodflowvelocity.PretreatmentwithGGDreversedtheoxytocinresponseonbloodflowvelocity.HistopathologicalexaminationshowedpretreatmentwithGGDalleviatedinflammationandedemaintheuteruswhencomparedwiththemodelgroup.BothexvivoandinvivoresultsindicatedthatGGDpossessedasignificantspasmolyticeffectonuterinetetaniccontractionaswellasimprovementonuterinearterybloodvelocitywhichmayinvolvePGF2αandCa2+signaling,suggestingthatGGDmayhaveaclinicpotentialinPDtherapy.

  • 简介:Gastrointestinaltoxicities(GIT),includingoralmucositis,nauseaandvomiting,anddiarrhea,arecommonsideeffectsofchemotherapyandtargetedagentsinpatientswithadvancedcolorectalcancerandpancreaticcancer.Beingoftenunderreported,itisstilldifficulttopreciselyestablishtheirburdenintermsofbothpatient’squalityoflifeandcancercarecosts.Moreover,withtheuseofmoreintensiveupfrontcombinationregimens,thefrequencyofthesetoxicitiesisrapidlygrowingwithapotentialnegativeeffectalsoonpatient’soutcome,asaresultofdosereductions,delaysorevendiscontinuationofactivetreatments.Thus,identifyingpatientsathigherriskofdevelopingGITaswellasanoptimalmanagementareparamountinordertoimprovepatient’scomplianceandoutcome.Afterthedescriptionofthemaintreatment-inducedGIT,wediscussthecurrentknowledgeonthepathophysiologyofthesesideeffectsandcommentthescalescommonlyusedtoassessandgradethem.WethenprovideacriticalupdateonGITincidencebasedontheresultsofkeyrandomizedtrialsconductedinpatientswithmetastaticcolorectalcancerandadvancedpancreaticcancer.
