简介:TheannualmeetingoftheHeartFailureAssociationofESCinLisbon,inJune2005,wasexceptionallysuccessful.Thereweremanyveryinterestingpresentationsandworkshopswiththeuniquetitle:StatinsinheartfailureCholesterol-loweringisnottheonlygoalHeartfailure(HF)isaprogressivediseasewithcoronaryarterydisease(CAD)asthemostoftenunderlyingetiology.TreatmenttopreventprogressionofheartfailurehasbeentargetedtoreversetheconsequencesofHFandtoalessextentthecause-theatheroscleroticplaqueitself.Ontheaverage50%ofpatientswithheartfailurearetreatedwithlipidintervention.Lipid-loweringtreatmentwithstatinsclearlyreducesmorbidityandmortalityofpatientswithdocumentedCAD.SincetheprevalentetiologyofheartfailureisCAD,itspreventionmayreduceheartfailureprogression.However,recentstudiessuggestthatpleiotropiceffectsofstatinsaremoreimportantthantheinfluencerelatedtotheircholesterolloweringmechanism.Furthermoreitissuggestedthatlowlevelsofcirculatinglipoproteinsandcholesterolmaybeindependentpredictorsofimpairedoutcomeinpatientswithheartfailure.Therearesomepossibleexplanationsforthisfinding.Highlevelsofcholesterolcanbebeneficialtoheartfailurepatients;cholesterol-richserumlipoproteinsareabletomodulateinflammatoryimmunefunctionbecausetheybindanddetoxifybacteriallipopolysaccharide,averystrongstimulatorofthereleaseofproinflammatorycytokinesthatpromoteheartfailureprogressionanddeath.SocurrentrecommendationsstronglyemphasizethattheaimoftreatmentofHFisnottolowercholesterol.
简介:ToelucidatethestructuresofSLA-DR(swineleukocyteantigenDR)genesofthreeChinesepigstrains(Gz,BmandYn),theSLA-DRAandSLA-DRBcDNAwereamplifiedbyRT-PCRandsubjectedtodeterminethesequences.ThewholestructuresofSLA-DRAallelesareidenticalamongthreestrains,consistingof759nucleotidesincludinganopenreadingframe(ORF),andaresharedwiththosereportedfromNIHminipigsSLA-DRAcandSLA-DRAd.ThesamelengthoftheORF-containingSLA-DRBgenesofthreeChinesepigstrainswasalsoidentified.Theyarecomposedof801nucleotidesencodingaxenogeneicantigenmoleculeof266aminoacidresidues.ThenucleotidesequencesoftheSLA-DRBgenes,however,aredifferentwhencomparedeitheramongthethreestrainsorwiththepublisheddataofSLA-DRBsequences,whichallowedournovelSLA-DRBallelesreceivingtheiraccessionnumbersAY102479,AY102480andAY102481fromtheGenBank.ThisstudyfurtherrevealsthatthephylogenichomologiesofMHCDRorDR-likegenesinstructuresofnucleotidesanddeducedaminoacidsbetweenChinesepigs(SLA)andhuman(HLA-DRB1*0901)arebetterthanthosebetweenpigsandmice(H-2bEβ).HighsimilaritieswerealsofoundforDRα-DRβheterodimersbetweenChinesepigsandhumanintermsofaminoacidssequencescriticalforbindingwithhumanCD4coreceptormolecule,whicharebetterthanthosebetweenSLA-DRandH-2I-Emolecules.Afunctionaltestindicatedthat,bycotransfectionwithBm-DRAandBm-DRBgenes,theBm-DRmolecule-expressedL929cellscouldstimulatehumanTcellsquitewellinaxenogeneicreactioninpresenceofhumanAPCs.
简介:Wntsignalingplaysanimportantroleinembryogenesisandtumorgenesis.AlthoughthemechanismabouthowWntstransducetheirsignalingfromreceptorfrizzled(Fz)tocytosolhasnotbeenunderstood,dishevelled(Dvl)proteinwasconsideredastheintersectionofWntsignaltraffic.Inthisstudy,wecharacterizedthefunctionofthreedomains(DIX,PDZandDEP)ofDvl-1incanonicalWntsignaltransductionandDvl-1membranetranslocation.ItwasfoundbothDIXandDEPdomainweresufficienttoblockWnt-3a-inducedLEF-1transcriptionalactivityandfreecytosolβ-cateninaccumulation;whereasPDZdomainandafunctionalmutantformofDEPdomain(DEP-KM)hadnoeffectoncanonicalWntsignaling.Inaddition,whencotransfectedwithFz-7,DEPdomain,butnotDIX,PDZorDEP-KM,translocatedandco-localizedwithFz-7totheplasmamembrane,whichwassimilartoDvl-1.Furthermore,itwasDEPdomainthatcouldblockFz-7-inducedmembranetranslocationofDvl-1viaapossiblecompetitivemechanism.TheseresultsstronglysuggestthatDEPdomainisresponsibleforthemembranetranslocationofDvl-1proteinuponWntsignalstimulation.

简介:ItiswellknownthatLandsatTMimagesarethemostwidelyusedremotesensingdatainvariousfields.Usually,ithas7differentelectromagneticspectrumbands,amongwhichthesixthonehasmuchlowergroundresolutioncomparedwiththeothersixbands.Nevertheless,itisusefulinthestudyofrockspectrumreflection,geo-thermalresourcesexploration,etc.ToimprovethegroundresolutionofTM6tothelevelasthatoftheothersixbandsisaproblem.Thispaperpresentsanalgorithmbasedonthecombinationofmulti-variateregressionmodelwithsemi-variogramfunctionwhichcanimprovethegroundresolutionofTM6by"fusing"thedataofothersixbands.Itincludesthefollowingmainsteps:(1)testingthecorrelationbetweenTM6andoneofTM1-5,7.IfthecorrelationcoefficientbetweenTM6andanotheroneisgreaterthanagiventhresholdvalue,thenselectthebandtotheregressionanalysisasanargument.(2)calculatingthesizeofthetemplatewindowwithinwhichsomeparametersneededbytheregressionmodelwillbecalculated;(3)replacingtheoriginalpixelvaluesofTM6bythoseobtainedbyregressionanalysis;(4)usingimageentropyasameasurementtoevaluatethequalityofthefusedimageofTM6.ThebasicmechanismofthealgorithmisdiscussedandtheVC++programforimplementingthisalgorithmisalsopresented.Asimpleapplicationexampleisgiveninthelastpartofthispaper,showingtheeffectivenessofthealgorithm.

简介:ObjectivesThelong-termbenefitoflatereperfusionofinfarct-relatedartery(IRA)afteracutemyocardialinfarction(AMI)iscontroversial,andthebenefitmechanismsremainuncertain.Lowdosedobutaminestressechocardiography(LDSE)canidentifyviablemyocardiumandpredictimprovementofwallmotionafterrevascularization.MethodsSixty-ninepatientswithfirstAMIwhodidnotreceivedearlyreperfusiontherapywerestudiedbyLDSEat5to10daysafterAMI.Wallmotionabnormalityandleftventricularsizeweremeasuredatthesametime.SuccessfulPCIweredoneinallpatientsat10to21daysafterAMIonset.Patientsweredividedintwogroupsbasedonthepresenceorabsenceofviablemyocardium.Echocardiographywasrepeatedsixmonthslater.ResultsTherewere157motionabnormalitysegments.89segments(57%)wereviableduringLDSE.26patients(38%)withviabilityand43(62%)without.Inviablegroup,leftventricularejectionfraction(LVEF)wasincreased(P<0.05),andleftventricularendsystolicvolumeindex(LVESVI)andwallmotionscore(WMS)weredecreased(P<0.05andP<0.01)significantlyat6monthscomparedwithbaseline.Butinpatientswithoutviability,LVEFwasdecreased(P<0.01),andLVESVIandleftventricularenddiastolicvolumeindex(LVEDVI)wereincreased(P<0.05)significantlyafter6months,andtheWMSdidnotchanged(P>0.05).LVEFincreased(P<0.05)andWMSdecreased(P<0.05)onLDSEduringacutephaseinpatientswithviability,buttheywerenotchangedinthenonviablegroup.ConclusionsLaterevascularizationofIRAinpatientswithpresenceofviablemyocardiumafterAMIisassociatedwithlong-termpreservationleftventricularfunctionandlessventricularremodeling.ImprovementofleftventricularsystolicfunctiononLDSEindicateslatephaserecoveryofleftventricularfunctionafterlaterevascularization.
简介:目的:探讨针刺内关与郄门穴对冠心病患者每博心输出量及左心室排血的峰值流速的影响。方法:60例冠心痛患者分为内关组和郄门组,分别在运针3min后和留针20min后测试每博心输出量,左心室排血的峰值流速。再与针刺前得测试结果相比较。结果:46例患者针刺3min后Q值和VP值较针刺前上升。留针20min后,34例患者Q值和VP值全部上升。结论:针刺内关和郄门可增强左心室壁的收缩能力,使心脏的每博输出量增加,改善冠心病患者的心肌缺血状态。
